Originally sequenced in 1971, myelin basic protein is believed to play a role in production and maintenance of the insulative myelin sheath around neurons.

Myelin is an opaque, white mixture of proteins, phospho-, and glycolipids that form an electrically insulating sheath surrounding axons of certain neurons, permitting signal propagation over extended distances. MBP is a highly variant protein component of myelin. Many isoforms have been identified, and most appear to play a major role in both construction and maintenance of the myelin sheath.

Damage to, or dysfunction of myelin basic protein, is widely implicated in many demyelinating diseases.

- Harauz, et al., 2004:Crystallography of MBP hints at a specific folding patterns necessary for proper formation of the myelin sheath, and suggests a role in multiple sclerosis.
- Popko et al., 1987: Mutant mice deficient in MBP display defects in myelination. Transgenic expression of MBP restores myelination, and results in partial recovery from symptoms of demyelination.
- Berger et al., 2003: Myelin autoantibodies are predictive of multiple sclerosis.
