We are delighted to announce the launch of our PolyUGTs® kits for synthesising glucuronides. Following innovative research by Hypha’s scientists with support from Innovate UK, we have successfully developed multiple UGT isoforms sourced from several of Hypha’s microbial biotransformation strains. The easy-to-use kits containing purified PolyUGT enzymes, enable scientists to make O-, N- and acyl glucuronides in their own labs, for:

  • Identification by NMR spectroscopy
  • Measurement of deconjugation of glucuronides during bioanalysis sample handling, and
  • Identifying routes to larger amounts (mg-g) of glucuronide material to support further studies
  • Lability testing and, for acyl glucuronides, measurement of acyl ring migration kinetics

The kits complement Hypha’s existing PolyCYPs kits used for making CYP and other phase I metabolites.

PolyUGTs have been tested for activity against a number of drug substrates known to be subject to glucuronidation. This includes glucuronidation of alkyl and phenolic hydroxyls for O-glucuronides, carboxylic acids for acyl-glucuronides, as well as aromatic nitrogen-containing systems for N-glucuronidation. Glucuronidation of some non-aromatic alkyl N-moieties is also observed.

Key features

  • Makes glucuronides for a wide range of substrate types

  • Comes with all reagents needed, including UDPGA co-factor, a control compound, pH reduction buffer (for acyl glucuronides), formulation reagent, and a 24-well reaction plate and seal

  •  Easy-to-use - all reagents are provided as lyophilised powders - just add water and gently shake

  • Readily scalable, either in-house or outsource to Hypha

Kits available

Screening kits – Allows you to identify which of 11 isoforms best produces your target glucuronide. Two parallel reactions are possible per kit.

Scale-up kits – Scale-up vials allow a repeat of screening reactions at x20 the reaction volume to produce material for MetID and biological testing.
